Basic Protocol 1: Growth of Streptomyces argillaceus/griseus in R5A Liquid Medium

  Materials
  • R5A liquid medium (see recipe)
  • Streptomyces argillaceus/griseus on solid medium or as a glycerol stock ( protocol 3)
  • 250‐ml baffled culture flasks, sterile (glass or plastic with vented caps)
  • Toothpicks, sterile
  • Sterile cotton, optional
  • Incubator/shaker apparatus

Basic Protocol 2: Growth of Streptomyces argillaceus/griseus on R5A Solid Medium

  Materials
  • R5A solid medium (see recipe) with appropriate antibiotic
  • Streptomyces argillaceus/griseus glycerol stock
  • 100 × 15–mm petri dishes
  • Sterile spreader
  • 28°C incubator

Basic Protocol 3: Long‐Term Storage of Streptomyces argillaceus/griseus

  Materials
  • Streptomyces argillaceus/griseus liquid culture
  • Sterile glycerol
  • 1.5‐ml microcentrifuge tubes
  • 1‐ml pipets
  • Pipettors
  • −80°C freezer
